서비스 절차
2-DE 분석 서비스를 위한 샘플 준비
- 세포: 배양된 세포를 2~3차례 PBS로 Washing 후 최종적으로 pellet만을 dry ice package로 보내주시면 됩니다.
필요한 세포의 양은 4 X 106 입니다. - 동물조직: 20mg의 조직을 dry ice package로 보내주시면 되고 blood가 많은 부분일 경우에 최대한 표면에 있는 것을 제거하여 보내주시면 됩니다.
- 식물조직: 식물조직은 채취 후 protease의 영향에 취약하므로 얼린 시료는 녹지 않도록 주의합니다.
액체질소에 고운 입자가 될 때까지 조직을 파쇄한 후 500mg 이상을 dry ice package로 보내주시면 됩니다. - IP샘플: 실험이 완료되면 sup을 모두 버리시고 bead만을 4℃ 상태를 유지하도록 ice pack을 넣어 보내시면 됩니다.
IP 샘플의 경우 사용하신 항체 유래의 단백질이 혼재되어 있기 때문에 샘플을 사용하지 않고 실험 전체 과정을 함께 진행한 Control sample이 대조군으로 필요합니다. - 배지나 기타 buffer 내의 단백질: 배지는 포함되어 있는 단백질 양이 제각각이므로 따로 권장량이 정해져 있지 않지만 100ml 이상을 보내주시길 권해드립니다. 기타 buffer 내에 있는 단백질인 경우 ice pack이나 dry ice에 넣어 보내주시면 됩니다.
- 기타 건조, 분말 샘플: 상온 상태로 보내시면 됩니다.
- 모든 샘플의 최소 단백질 양은 SILVER staining일 경우 300㎍ 이상, CBB staining을 원하실 경우 800㎍ 이상을 보내주셔야 합니다. 다만 IP샘플이나 기타 최종 분석하고자 하시고자 하는 시료의 단백질 양이 이에 못 미칠 경우에도 2-DE분석은 가능하지만 추후 MS 분석에서 동정 성공률에 영향을 끼칠 수 있음을 미리 알려드립니다.
2DE 분석에 이어 이미지 분석에 따른 발현변화를 ±0.1 fold까지 분석하고 이중 선정된 또는 지정하신 spot에 대해 package 분류에 따라 일정 개수의 spot을 동정분석 하는 package 서비스입니다.
